Colegio de Sta. Rosa Makati traces its roots to its mother school, Colegio de Sta. Rosa Intramuros. The institution was founded in 1750 by M. Paula de la Santisima Trinidad, a Dominican Tertiary from Spain. The school buildings of the campus in Intramuros were destroyed during World War II but was reconstructed in 1948. In 1964, the Congregation of the Servants of St. Joseph, inspired by the institution's founder, responded to the demands for a Catholic institution in the municipality of Makati. Colegio de Sta. Rosa acquired a lot offered by the Makati Development Corporation, under the leadership of Rev. Fr. Moises Lopez, Order of Augustinian Recollect (OAR). Soon after, the institution received government permit to open Colegio de Sta. Rosa, as a branch of CSR, Intramuros. The school was officially opened in 1964. Colegio de Sta. Rosa holds Level 2 accreditations in both their Grade School and High School Departments awarded by the Philippine Accrediting Association of School Colleges and Universities (PAASCU).
